What constitutes "nighttime hours" for the subway? I'm trying to figure out the best way to get to/from Brooklyn Masonic Temple...
I'll think more, Matt, but you should go to the Brooklyn Brewery for a tour. They have them Saturdays and Sundays for free at certain times throughout the day. I haven't been yet, so I can't attest to its quality, but I do like their beers.
I think after 12 is nighttime hours. Sometimes, before that, I'll get on the 2 and 3 and they'll already be running local though. It should be easy to go to Brooklyn. You won't be on nighttime hours yet. Afterwards, when you are leaving Brooklyn, the trains might just be making all the local stops (and it might take a while for a train to show up). The Masonic Temple is not a long walk from the A, C, 2, 3, 4, 5 - those trains can take you anywhere in Manhattan, pretty much.
Also, if you somehow end up in a part of Brooklyn where there are L-Stations like I did last Saturday, you will be disappointed to find that some of them have no trains to Manhattan at night, and you'll have to journey toward Williamsburg until you find one.

For the East Village I recommend TenEleven at 11th Street and Avenue C. Good jukebox, good alcohol selections. And when we got sick of the jukebox the people at the bar let me run a playlist from my Droid. They have nights where they play cult/weird movies on the projector in the back room.
For Brooklyn I recommend Fette Sau for grilled meats, beer and bourbon. The nearby GoodCo is fun, too, and if you go there Sunday nights Nando (who posts sometimes on these boards) DJs a karaoke night which is very fun.
Barcade can get crowded by is also fun. They have a ton of great beers on tap and more arcade video games than you could possibly play.
